Kersen leaves contain secondary metabolites, alkaloid, flavonoid, tannin, saponin, steroids. High flavonoid compounds require toxicity testing. This study aimed to determine the acute toxicity of ethanol fraction of Mutingia calabura L. leaves in vivo on Wistar strain male white rats by using fixed dose procedure method. In this study, 2 tests were carried out, the preliminary test and the main test (2 groups, normal & treatment at a dose of 2,000 mg/KgBW).The result of total flavonoid content in the ethanol fraction of kersen leaves is 139,50 mg QE/g or 13,95%. The result of observation in the main test showed that were no deaths or toxic symtomps in the test animal. Average level of biochemical parameters of normal group are SGOT 135,203 ? 20,569 U/L, SGPT 86,372 ? 12,604 U/L, creatinine 0,604 ? 0,062 mg/dL, ureum 38,73 ? 4,176 mg/dL whereas the dosage group of 2.000 mg/KgBB are SGOT 127,752 ? 20,376 U/L, SGPT 75,576 ? 8,406 U/L, creatinine 0,55 ? 0,0616 mg/dL, ureum 36,916 ? 2,257 mg/dL. A single dose of the ethanol fraction of cherry leaves at a dose of 2,000 mg/KgBW had no effect on the biochemical parameters of liver, heart, kidney and macroscopic functions of the liver, kidney and heart.} }
