Abstract :
ABSTRACT ANALYSIS OF DRUG CHEMICAL PARACETAMOL CONTAMINATION IN TRADITIONAL MEDICINE STIFF PAIN USING THIN LAYER CHROMATOGRAPHY (TLC) AND LIQUID CHROMATOGRAPHY-MASS SPECTROMETRY (LC-MS) Rini Setyowati Analgesic drugs and NSAIDs are often add illegally in traditional medicine stiff pain like paracetamol, so that the possibillity of paracetamol as one of the drugs added were very larger. This purposes of this research were to know drug chemical contamination of paracetamol and concentration of paracetamol in traditional medicine stiff pain in the area of Borobudur District. This research is an experimental research, for the data analysis using microsoft excel and linear regression. The qualitative test method was use the organoleptic test, Thin Layer Chromatography (TLC) test with the eluent ratio of chloroform : ethanol (9 : 1) and silica gel GF254 as a stationary phase, and Fourier Transform Infrared Spectroscopy (FTIR) test. The quantitative test method uses the Liquid Chromatography-Mass Spectrometry (LC-MS) test with the eluent ratio of A = H2O + 0,1% formid acid : B = methanol p.a + 0,1% formid acid (A : B = 5 : 9) and stationary phase is the detector. The result identification of the organoleptic test were the shape, color, smell, and taste of the sample traditional medicine stiff pain. The results of the TLC test on sample A and B were positive, indicated by a purple spot and the Rf value of the sample compared to standard Rf value of paracetamol, while sample C is negative. The result of the FTIR test on sample A and B were positive because the functional group is almost the same as the standard paracetamol functional group, while sample C is negative. LC-MS test results after calculating the concentration in samples A and C the concentration were obtained kurang lebih 4,4 ?g/kg below the Detection Limit (LoD) : 4,4 ?g/kg, while sample B the concentration were obtained 2,22%. Based on the results of all three samples tested, the sample A and sample B were positive contamination by drug medicine paracetamol while sample C were negative.
