Abstract :
Fixation is one of the histotechnical stages that aims to inhibit the process of
decay, autolysis, preserving or maintaining the shape of cells that exist in the
tissue to remain intact, colloidal solidification and effect on staining. The purpose
of this study was to determine fixation fluids with better penetration power to the
quality of microscopic images in tissues after the staining of Hematoxylin-Eosin
(HE).
This study uses a type of literature study by collecting reference data
obtained from research results relating to differences in fluid fixation in tissues
with Hematoxylin-Eosin (HE) staining such as fixative fluids Neutral Buffer
Formalin (NBF) 10%, Alcohol 70%, Alcohol, Bouin and Methanol with research
sample is liver tissue. The data obtained were then analyzed using descriptive
analysis.
The results obtained, that the quality of a good tissue preparation for
fixation is the type of 10% NBF solution and Bouin which results show the
quality of the preparation was observed and an assessment of score 3 with good
results that appear blue in the cell nucleus, red color in the cytoplasm is clear and
the colors in the preparations are uniform, whereas in tissues fixed with 70%
Alcohol fixation fluid, Acetone and Methanol with a score of score 2 with
unfavorable results that show the results of blue in the cell nucleus less, red in the
cytoplasm as well as colors in the preparations less uniform.
