Abstract :
Celery leaves as one of the plants that are widely used by people as a food
flavoring. Celery leaves can be used as antihypertensive, anti-rheumatic, and
reduce uric acid, cause the celery leaves contains flavonoid compounds. This
study aims to determine the total flavonoid levels and antioxidant activity of the
etanol extract, n-hexane fraction, ethyl acetate fraction, and water fraction. In the research, the ethanol extract of celery leaves (Apium graveolens L.)
was obtained by maceration using 70% ethanol for 5 days and remaceration for 2
days, and then fractionated by liquid extraction method using separating funnel.
The antioxidant activity was determined using ABTS radicals. Ethanol extract, n-hexane fraction, ethyl acetate fraction, and water
fractions had total flavonoid that equivalent of quersetin were 63.16%, 83.39%,
88.89%, 67.55% respectively. Antioxidant activity was expressed as IC50 values in
the extract, the n-hexane fraction, the ethyl acetate fraction, and water fraction
were 24.63?0.23 ppm, 17.35?0.22 ppm, 15.50?0.14 ppm, 17.66?0.28 ppm
respectively. Based on the result obtained, the antioxidant potency of extract and
fraction were successively of ethyl acetate fraction ? n-hexane fraction ? water
fraction ? extract. Statistical analysis using one way ANOVA obtained a
significant value of p?0.05. The result indicate there are differences in total
flavonoid content and antioxidant activity between ethanol extract and fractions
from celery leaf.
