Abstract :
Papaya seeds (Carica papaya L.) contain flavonoids, phenols, saponins, and alkaloids that have the potential to act as natural antioxidants. The papaya
seed ethanol extract which has antioxidant activity is formulated in the form of a
gel dosage. The purpose of making a gel preparation is because it has good drug
release and good ability to spread on the skin. In addition, to determine the
optimal combination of HPMC and Karbopol concentrations in papaya seed
ethanol extract gel which can provide antioxidant activity. In this study, using maceration methods and Simplex Latice Design on the
preparation formula to obtain the optimum formula based on the parameters of the
pH test, spreadability, adhesion, and viscosity. Verification of the optimal formula
used is the statistical test using the one sample t test. In measuring antioxidants
using the ABTS method (2,2'-azino-bis (3-ethylbenzothiazolin) -6-sulfonic acid)
which is expressed in the form of IC50 values. The results showed that the yield of papaya seed ethanol extract was
17.7%. The optimum formula for the gel preparation contains a papaya seed
ethanol extract of 1.6%, based on the Simplex Latice Design method, namely on
the basis combination of HPMC 4.96 grams and 0.04 grams Karbopol with a
desirability value of 0.952. Based on the pH test value of 5 ± 0.00, the
spreadability test of 5.37 ± 0.28, the adhesion test of 2.47 ± 0.13, and the viscosity
test of 600 ± 0.00. The test value of each physical characteristic is declared valid. Based on the one sample t test and the results of the optimum cycling test, the
formula is stable. The average IC50 value of the antioxidant activity of the papaya
seed ethanol extract gel was 34.99 ± 0.065 ppm with a very strong category.
