Abstract :
Kepok banana weevils contain flavonoid compounds that can be used as
antibacterial agents. This study aims to prove the ethanol extract and fractions from
the ethanol extract of kepok banana hump in inhibiting Escherchia coli ESBL
(Extended Spectrum Beta Lactamase) bacteria and comparing its inhibitory ability
with Chloramphenicol positive control.
Kepok banana weevils were macerated with 96% ethanol then dissolved in
warm water and fractionated using n-hexane and ethyl acetate solvents.
Antibacterial testing was carried out on the extract, n-hexane fraction, ethyl acetate
fraction and water fraction of Kepok banana hump. The method used for testing
antibacterial activity is the diffusion method. The negative control used DMSO
10%, while the positive control used was chloramphenicol. The results are shown
from the radical inhibition zone diameter. Antibacterial statistical analysis used one
way ANOVA.
The radical inhibition zone of ethanol extract with a concentration of 25%
was 6.66 mm, a concentration of 50% was 7 mm, while for a concentration of 100%
was 6 mm. The n-hexane fraction at concentrations of 25%, 50% and 100% gave
the same radical zone, namely 6 mm. Ethyl acetate fraction with 25% concentration
of radical inhibition zone obtained was 16.95 mm, 50% concentration was 13.3 mm
while at 100% concentration was 11.16 mm. The concentration of water fraction
25% and 50% obtained 6 mm radical zone, 100% concentration obtained a radical
inhibition zone of 6.33 mm. Ethyl acetate fraction with 25% concentration gave a
bigger radical inhibition zone against Escherichia coli ESBL, which was 16.95 mm
including intermediates, not yet equivalent to the positive control chloramphenicol
which had a radical inhibition zone of 31.9 mm (sensitive). The data also supported
statistically there was a significant difference (p <0.05) between the ethyl acetate
and chloramphenicol fractions.
