Abstract :
The use of kluwih plants so far has only been focused on the fruit and stems,
while the leaves of kluwih (Artocarpus camansi) also contain many active
compounds, especially flavonoids. The purpose of this study was to determine the
total flavonoid levels and antioxidant activity of the extract and fraction of kluwih
leaves (Artocarpus camansi).
The extract was made by maceration method using 70% ethanol solvent
followed by the addition of warm water then fractionation using n-hexane and ethyl
acetate solvents. Determination of flavonoid levels and antioxidant activity was
carried out using a UV-Vis spectrophotometer. The antioxidant activity was based
on the free radical scavenging ability of 2,2'-azinobis (3-ethylbenzthiazoline 6-
sulfonic acid) (ABTS) compounds measured at a wavelength of 427.5 nm at 2
minutes. The analysis was conducted to determine the differences in total flavonoid
levels and antioxidant activity between extracts and fraction of kluwih leaves
(Artocarpus camansi) using One Way ANOVA.
In phytochemical screening, extracts and fractions of kluwih leaves contain
flavonoids. The results of the determination of the total flavonoid mean content in
the ethanol extract were 2.9950 ± 0.0196% QE, the n-hexane fraction was 1.0262
± 0.0175% QE, the ethyl acetate fraction was 2.5019 ± 0.0468% QE , and the water
fraction of 1.5165 ± 0.0270% QE. The average antioxidant activity is expressed in
IC50 values with the results in the ethanol extract of 16.7181 ± 0.2318 ppm, the nhexane fraction 23.0089 ± 0.3990 ppm, the ethyl acetate fraction 18.2579 ± 0.2936
ppm , and the water fraction of 18.5776 ± 0.0697 ppm. Based on one way ANOVA
analysis on total flavonoid levels, there was a significant difference between ethanol
extract and kluwih leaf fraction (Artocarpus camansi) (p <0.05). There was a
significant difference in antioxidant activity between ethanol extract and kluwih
leaf fraction (Artocarpus camansi) (p <0.05).
