Abstract :
TSIA is a differential medium with a pH indicator that can distinguish
microorganisms based on their ability to decompose specific carbohydrates,
namely glucose and lactose or sucrose with or without producing gas. The media
used has two parts, namely slant and butt. Changes experienced after incubation,
namely the color of the medium yellow indicates acid, the color of the medium
becomes redder indicates the medium becomes alkaline. The aims of this study
was to determine the reading time of the results of the formation of fermentation
on TSIA media by several gram-negative bacteria at various incubation times.
This study used a descriptive observational design. This study was
conducted at the STIKES Nasional Bacteriology Laboratory and the time of the
study was carried out in January-May. The samples of this study were bacterial
cultures of Escherichia coli, Klebsiella pneumoniae, Serratia marcescene, and
Providencia rettgeri, isolated media of TSIA (Triple Sugar Iron Agar) which were
incubated with variations in incubation time of 8, 16, 24, and 32 hours. The
sampling technique used is Quota Sampling.
The results of study showed that results of acid/acid in Escherichia coli and
Klebsiella pneumoniae bacteria within an incubation time of 8 hours fermenting

/4 parts slant and butt. For alkaline/acid results in Serratia marcescene, the acidic
part of the butt was fermented 4
/4 parts within an incubation time of 8 hours, while
for Providencia rettgeri bacteria the acidic part of the butt was fermented 3
/4 parts
at an incubation time of 8 hours.
Based on the results of study that has been carried out regarding the
observation test for the formation of fermented products on TSIA media at various
incubation times, it can be concluded that there are differences in the results of
acid/acid and alkaline/acid fermentation on TSIA media produced by bacteria
Escherichia coli, Klebsiella pneumoniae, Serratia marcescene and Providencia
rettgeri at various incubation times of 8, 16, 24, and 32 hours.
