Abstract :
Red dragon fruit peel (Hylocereus polyrhizus) of content flavonoid
compound that are beneficialas antibacterial. The research determine of total
flavonoid content and antibacterial activities of extracts ethanol and fractions
ethyl acetate red dragon fruit peel against Escherichia coli ans Staphylococcus
aureus.
Red dragon fruit peel of extracted using 70% ethanol solvent of
maceration method and fractionatedusing n-hexane and ethyl acetat solvent.
Determination of flavonoids content using aluminium chloride method with UVVis spectrophotometry and the results were analyzed using the Independent
Sample T-Test. Antibacterial activities using the diffusion disk method with
concentrations of 25%, 50% and 75%, using negative control a DMSO 10% and
positive control a Ciprofoxacin.
The result of identification flavonoids compounds indicate red dragon
fruit peel extracts ethanol and fractions ethyl acetat contain flavonoids
compounds. The determination of total flavonoid contained in extracts ethanol
and fraction ethyl acetat of 8,773 %QE ± 0,122 and 11,596 %QE ± 0,089. There
was a significant difference in the average total flavonoid content in 70% ethanol
extract with the ethyl acetate fraction of red dragon fruit peel (Hylocereus
polyrhizus) (p<0.05). The average results of the antibacterial inhibition zones of
the ethanol extract of red dragon fruit peel with concentrations of 25%, 50% and
75% against Escherichia coli bacteria were 7.3 mm, 8.3 mm and 9.5 mm, while
the ethyl acetate fraction of red dragon fruit peel concentrations of 25%, 50% and
75% against Escherichia coli bacteria obtained an average of 8 mm, 9.5 mm and
11.2 mm. The average results obtained on the ethanol extract of red dragon fruit
peel with concentrations of 25%, 50% and 75% against Staphylococcus aureus
bacteria were 7.6 mm, 8.6 mm, 9.8 mm, while for the zone average results Inhibit
the ethyl acetate fraction of red dragon fruit peel against Staphylococcus aureus at
concentrations of 25%, 50% and 75%, namely 8 mm, 9.4 mm and 11.9 mm.
Results Based on the Post Hoc Test, the ethanol extract and the ethyl acetate
fraction of red dragon fruit peel had antibacterial activity against Escherichia coli
and Staphylococcus aureus because the zone of inhibition was significantly
different (p < 0.05) with the negative control (DMSO 10%). The results of the
inhibition zone obtained by the ethanol extract and the red dragon fruit peel
fraction were not equivalent to the inhibition zone obtained in the positive control
of Ciprofloxacin against Escherichia coli and Staphylococcus aureus bacteria,
namely 36 mm and 26.66 mm
