Abstract :
Teratai leaf (Nelumbo nucifera Gaertn.) has_a biological activity as an antiinflammatory, anti-cholesterol and antidiabetes, The leaves contain alkaloids, flavonoids, tannins and triterpenoid/steroid. The purpose of this study was to determine n-hexane extract (ENHDT), ethylacetate extract (EEADT) and ethanol extract (EDT) teratai leaf that can lower blood glucose levels in mice induced by streptozotocin and then the most active extract were analyzed by paper chromatography (KKt) and thin layer chromatography (TLC), Simplicia powder was extracted with a different solvent polarity (n- hexane, ethyl acetate and ethanol). Diabetic mice (n = 58) were randomly divided into 11 groups, control group treated with 0.5% carboxymethyleellulase(CMC) suspension, positive control group treated with metformin suspension dose 63 mg/kg bw and the test group were given ENHDT, EEADT and EEDT extract with a dose of 50 mg/kg bw, 100 mg/kg bw and 150 mg/kg bw. Treatments of mices conducted for 15 days, the results then analyzed with ANOVA and Duncan methods. The characterization of simplicia result water content of 8.65%; water soluble extract concentration of 17.65%; ethanol soluble extract concentration of 16.23% total ash content of 7.2%; and insoluble ash content in acid of 1.69%; total bacteria contamination (-) negativ colony/g and total mold and yeasts contamination (-) negativ colonvig. The most active extracts can lower blood glucose level in mice is EEADT.EEADT at dosages of 100 mg/kg bw hasnot significantly different with metformin 65 mg/kg bw. Chromatogram of BEADT in paper chromatography result 4 flavonoids spots green (Rf = 0.79) yellow (RE = 0.6); green yellow (RF = 0.38); and blue (RF ~ 0.13) with AICls 3% in mobile phase BAW (4:1:5). Thin layer chromatography result 3 spots blue-black phenolic compounds (RF= 0.92; RF= 0.8; RF= 0.78) with FeCl3% in ethylasetat : methanol (7:3).
