UJI AKTIVITAS ANTI-INFLAMASI DAN PEMBUATAN SEDIAAN ROLLER GEL PIPERIN TERHADAP PENGUKURAN EDEMA DAN JUMLAH NEUTROFIL PADA PUNGGUNG MENCIT YANG DIINDUKSI KARAGENAN

Abstract :
Piperin merupakan senyawa golongan alkaloid yang mampu menekan respon inflamasi. Tujuan penelitian ini untuk memperoleh sediaan roller gel sebagai anti- inflamasi. Sediaan roller gel dibuat menjadi 3 variasi formula, yang membedakan pada setiap formula yaitu dosis piperin yang digunakan. Formula 1 mengandung piperin 5 mg, formula 2 mengandung piperin 10 mg dan formula 3 mengandung piperin 15 mg. Pengujian menggunakan hewan mencit Swiss Webster jantan. Mencit dibagi menjadi 6 kelompok uji, terdiri dari kelompok normal, kontrol negatif, kontrol positif, formula 1, formula 2 dan formula 3. Semua kelompok hewan, bagian kulit punggung diinduksi dengan karagenan 3%. Parameter aktivitas anti-inflamasi adalah melihat penurunan edema dan penurunan rerata jumlah neutrofil pada kulit punggung mencit. Hasil persentase inhibisi pengukuran tebal lipat punggung mencit pada formula 1 sebesar 20,42%, formula 2 sebesar 28,24% dan formula 3 sebesar 20,24%. Hasil evaluasi rerata penurunan jumlah neutrofil formula 1 sebesar 26, formula 2 sebesar 19,6 dan formula 3 sebesar 41,6. Formula yang paling baik dari segi fisik dan penghambatan respon inflamasi ditunjukkan oleh formula 2.;---Piperin is an alkaloid type compound capable of suppressing inflammatory responses. The purpose of this research is to acquire a gel as an anti- inflammatory dosage. The preparation of the gel roller is made into 3 variations of formula, which distinguishes each formula i.e. the dose of piperine used. Formula 1 contains a 5 mg Piperine, Formula 2 contains a piperine of 10 mg and Formula 3 contains Piperine 15 mg. Testing the use of an animal mice Swiss Webster. Mice are divided into 6 test groups, consisting of normal groups, negative controls, positive controls, Formula 1, Formula 2 and Formula 3. All animal groups, the back skin section is induced with a 3% carrageenan. The Parameter of anti-inflammatory activity is to see decreased edema and decreased average neutrophil count on the squeaky back skin. The result of the inhibition percentage of the thickness of the back folding measurement of Formula 1 is 20.42%, Formula 2 is 28.24% and Formula 3 is 20.24%. The evaluation result averaged a decrease in the number of neutrophils in formula 1 is 26, formula 2 is 19.6 and formula 3 is 41.6. Formula 2 was the best of physical performance and inhibition of the inflammatory response.
